Proglycem (diazoxide) is non-diuretic benzothiadiazide used to suppress insulin secretion in hyperinsulinemic hypoglycemia. Activates pancreatic β-cell K-ATP channels, hyperpolarizing membrane and reducing insulin release.

How Proglycem Works:

Hyperinsulinism causes inappropriate insulin secretion and severe hypoglycemia — inoperable insulinoma, congenital hyperinsulinism (CHI), nesidioblastosis, post-bariatric hypoglycemia.

Diazoxide opens K-ATP channels in β-cells — opposes insulin secretion. Effective for most causes but congenital hyperinsulinism from certain K-ATP gene mutations may not respond.

Proglycem oral suspension 3–8 mg/kg/day in 2–3 divided doses. Adjust to glucose response.
