How Lioresal Works:
Spasticity occurs when upper motor neuron lesions cause spinal reflexes to become disinhibited. Baclofen functions as a GABA-B receptor agonist that lowers muscle tone and spasms by inhibiting monosynaptic and polysynaptic reflexes directly at the spinal cord level. Delivering the medication intrathecally bypasses the blood-brain barrier to concentrate the drug exactly where it is needed. This targeted approach provides a much stronger antispasticity effect than oral baclofen while significantly reducing overall systemic drug exposure.
Form and use:
Lioresal Intrathecal is continuously administered using an implanted, programmable pump system, such as the Medtronic SynchroMed II. Before a patient can undergo pump implantation, they must first receive an initial test dose delivered via a lumbar puncture. Once the device is successfully implanted, the daily dosage is carefully titrated based on the patient’s specific clinical response. To maintain ongoing therapy, a trained nurse or physician must refill the pump every one to six months using a needle inserted through the skin.
