Depo-Estradiol is long-acting injectable estradiol — typically every 3–4 weeks IM injection. Used for menopausal vasomotor symptoms, hypogonadism, and other indications including off-label use in gender-affirming hormone therapy (feminizing).
How Denavir Works:
Estradiol is the primary biologically active estrogen. Estradiol cypionate is the esterified form — slowly released from oil-based IM depot over weeks. Reduces frequency of administration vs daily oral or weekly transdermal.
Indications: relief of menopausal symptoms (hot flashes, vasomotor symptoms), hypoestrogenism replacement, gender-affirming hormone therapy (off-label). Risks: venous thromboembolism, breast cancer, stroke, MI — similar to other estrogen routes but oral has higher VTE risk than transdermal/injection.
Form and use:
Depo-Estradiol IM injection every 3–4 weeks. Dose individualized.

BOXED WARNING for endometrial cancer (unopposed estrogen in women with uterus — add progestin), breast cancer (combination HRT especially), cardiovascular disease (stroke, MI, VTE/DVT/PE), probable dementia (in older postmenopausal women). Other warnings: gallbladder disease, hypertriglyceridemia, hepatic impairment, hypocalcemia, severe hypocalcemia, retinal vascular thrombosis, malignancies.
