Glasdegib selectively inhibits Smoothened (SMO) to block this pathway and target these resistant stem cells. When combined with low-dose cytarabine (LDAC), it has been shown to improve overall survival in unfit AML patients compared to LDAC alone. However, this specific regimen has been largely superseded by more effective venetoclax-based combinations.

Form and use:

Daurismo is administered as a 100 mg oral tablet taken once daily, and it can be consumed with or without food. It is taken in tandem with concomitant LDAC, which is given as a 20 mg subcutaneous injection twice daily for 10 days of every 28-day cycle. This combination therapy is continued until the disease progresses or unacceptable toxicity occurs. Patients undergoing this treatment require regular ECG and electrolyte monitoring due to the risk of QT prolongation.

Generic availability:

As of 2026, there is no generic version of Daurismo available in the United States. For unfit AML patients, preferred alternative treatments include venetoclax combined with either azacitidine or decitabine due to their strong clinical outcomes. Depending on the patient’s specific AML genetics, other targeted oral options are also utilized. These include IDH1/IDH2 inhibitors like Tibsovo and Idhifa, as well as FLT3 inhibitors like Xospata and Rydapt.

Daurismo carries significant warnings for QT prolongation, requiring electrolyte correction and the avoidance of other QT-prolonging drugs.
